I guess that depends on what you consider a city. The first European settlement in what is now Arizona was the strong Spanish in Tubac (south of Tucson), which was founded in 1752. This was a military fortress walls (or prison), but a small settler community formed around him. A few decades before that, there were Spanish missions founded in San Javier (south of Tucson) and Guevavi (near the border with Mexico) that usually had some families Spanish nearby, besides the priests, but not really people called in the traditional sense. In 1776, the garrison at Tubac was transferred to a new location in Tucson, where the surrounding community eventually grew to become the biggest strength than their own and have streets, shops, church and other partners to a city true.
